Antibiotic abuse, ban antibiotics use, how can we do? Good antibiotics substitutes - lysozyme

Because of the abuse of antibiotics, there are many side effect appeared:
1. More and more Drug resistant bacteria, such as Common bacteria like salmonella, E. coli and Methicillin-resistant Staphylococcus aureus (MRSA), threatening our ability to treat illness and disease in humans
2. Drug residues harmful to human health
3. Kidney, liver toxicity effect animal health growth

The European Union, Korea, USA, Canada have prohibited the antibiotics use in feed.
China and Brazil have prohibited the use of colistin sulfate in feed

Decision of the four session of the twelve National People's congress:
1. Prohibit use lomefloxacin, pefloxacin, norfloxacin and ofloxacin for food animal from 31st,Dec 2016;
2. Intends to prohibit colistin sulfate use as growth promoter, recently are comments to the community, is expected to be completed by the end of 2016;
3.  Encourage and support the development of new veterinary drugs, such as microbial, antimicrobial peptides, herbal extracts, to improve the level of animal health, reduce the use of antibiotics as growth promoter and the use of veterinary drugs

With the forbidden of colistin sulfate, II antibiotics and olaquindox, the feed additives to against the gram-negative bacterial almost none available, how to solve the weaning piglets diarrhea problem is the top priority of feed companies, seeking effective antibiotics alternatives is extremely urgent and important.

Microbial lysozyme is the best choice, lysozyme is a protein which could dissolves bacteria, named lysozyme (muramidase).

Rumor has it that in 1921, a droplet from Fleming’s nose (who was nursing a cold while working in his lab) accidentally fell into one of the Petri dishes containing microorganisms. Noticing the reaction of the bacteria to the fluid, Fleming deduced that it must have contained a “remarkable bacteriolytic element”, and so he set out to isolate it.
That same year he announced that this element, named LYSOZYME because of its lysing antibacterial action.

Lysozyme is divided into different types according to the source, there are animal & human lysozyme, plant lysozyme, microbes lysozyme and egg-white lysozyme. Now, the most common lysozyme in the market is egg white lysozyme which is extracted from eggs, it is widely used in food industrial as preservative. Microbial lysozyme are fermented lysozyme.

Microbial lysozyme has very good bactericidal effect on gram-positive bacteria and gram-negative bacteria in a low concentration, it could be verified by tests.

Brief of Aegis lysozyme

Application

Using Case Sharing:

1. Big pig farm in Zhejiang province

(1)Program:
I. Control group was fed by normal basal diet, creep feed (DBN S100), piglet feed (40% of the Guangzhou 8V premix) 
II. The test group added microbial lysozyme 10% into the free-antibiotics ration according to 1 kg/Ton ( microbial lysozyme supplied by Zhejiang Aegis Biotech Co. Ltd.)
III. Experimental pigs: three-crossbred Pigs, the average age was 26 days.

(2) Result of creep feed. Test period: 8 days
 ADW increased 22.7g
 ADFI increased 49.35g

(3) Result of nursery pig. Test period: 19 days

ADW increased 24g
ADFI increased 22.4g
FCR decreased 0.064

During the test, there were 10 pigs diarrhea of control group.None diarrhea of lysozyme group.

Several common pig diarrhea diseases

1. Yellow scour of newborn piglets
Caused by Escherichia coli, multiple occurred in piglets 1 weeks after the birth, most common on 1~3 days age .
Clinical manifestation: normal when birth, then start diarrhea, stool was yellow, paste, short course, quick death.

2. White scour of piglets
Caused by Escherichia coli, multiple occurred in 10~30 days piglets, most common on 2~3 weeks age .
Clinical manifestation: piglets diarrhea suddenly, began to row of stool like paste, and then like water, followed by a white, gray or yellow white diarrhea, have fishy smell, pigs weight loss rapid, the mortality was lower.

3.Red dysentery
Caused by Clostridium welchii, multiple occurred in 3 days piglets.
Clinical manifestation: row red-brown or gray stench loose stool, accompanied by vomiting. Pigs quick death, high mortality.

4. Piglet paratyphoid
Caused by Salmonella, multiple occurred in 6 month younger piglets. most common on 1~4 month age .
Clinical manifestation: water samples loose stool, vomiting, shortness of breath, there are skin purpura on ears, limbs, lower abdomen.

5. Transmissible gastroenteritis
Caused by a coronavirus, all ages pigs were susceptible to the virus, but it was the most common in 10 days younger age piglets.
Clinical manifestation: sudden vomiting, water samples loose stool, yellow green or gray feces, rapid weight loss and dehydration, usually death after 2 to 7 days infection of illness.

Test Purpose:
To verify that microbial lysozyme could help pigs growth faster, less diseases, and reduce mortality rate and elimination rate.

Test program:
Because test is on weak weaned piglets, use creep feed.

LYZ- lysozyme - microbial lysozyme

Lysozyme History

Lysozyme, a remarkable natural antimicrobial and antiviral enzyme, discovered by Alexander Fleming in 1921, when a droplet from Fleming’s nose (who was nursing a cold while working in his lab) accidentally fell into one of the Petri dishes containing microorganisms, noticing the bacteria reaction, Fleming set out to isolate it and find a dissolving bacterial  protein, he named it lysozyme.

Lysozyme Introduction

The lysozyme is a kind of Glycoside hydrolases which is widely present in the plant and animal. It mainly acts on the ß-1,4 glycosidic bond between NAM and NAC. The lytic mechanism for bacterial killing by lysozyme include enzymic peptidoglycan hydrolysis of bacterial wall and induction of autolysis that can cause bacterial autolysis.



As a nonspecific immune factors in body fluids and organizaiton, lysozyme participates in kinds of immune response to improve and enhance the swallowing & digestive function of macrophages, and activate sallowing function of other interleukin, then enhance immunity. It can play an important rule in maintaining the intestinal flora balance, promoting the Bifidobacterium proliferation, prevention of enteritis and allergy.In addition, lysozyme also can can directly act on the viral protein with loads, to make the virus inactivation.

Lysozyme Classification

Lysozyme could be divided into different types according to the source: Egg-white lysozyme, Human and mammalian lysozyme, plant derived lysozyme and microbial lysozyme.
At present, the most common is the egg white lysozyme, which is widely used in food and feed industry. But there is an obvious defect - it did not showed satisfactory on gram-negative bacteria.

Probiotics can improve the growth performance and immune function of animals by establishing the dominant bacteria group, promoting the nutrients absorption and producing antibacterial substances. Immune polysaccharide could promote the growth of beneficial bacteria, inhibit the growth of pathogenic bacteria, improve the growth performance of animals mainly by improve animal digestive tract flora.
   As a non-specific immune factor, lysozyme is involved in many kinds of immune responses, improving and enhancing the phagocytosis and digestion of macrophages, enhancing the body's resistance to maintain the balance of intestinal flora and promoting the growth of animals.
   This test mainly study the effects on growth performance and immune function in broilers of lysozyme, probiotics and Astragalus Polysaccharide.
1 materials and methods
1.1 Test materials
   Lysozyme was provided by Zhejiang Aegis Biotech Co., Ltd; Lactobacillus and Bacillus were provided by Xi'an Canghai Biological Co., Ltd; Aastragalan was provided by Sichuan Deyang city Hoycome Bio-Tech Co., Ltd.
  1. 2 Testing animal & designation
   Select 288 1 day old healthy Avian broilers, divided into six groups randomly, each group have four repeat, 12 chickens in each replicate. Feed them ration, 100g/t lysozyme supplement ration, 5 billion CFU/g probiotics supplement ration and 320 g/t astragalan supplement ration.
   The test ration were formulated with corn and soybean meal, and the nutrition level was shown in Sheet 1.

  2. 3. 1 Determination of growth performance
   Weighting the 1st, 21st and 42nd days broiler, calculate the feed consumption, the daily average weight gain, average daily consumption and feed conversion rate of each stage.
  1. 3. 2 Determination of serum biochemical parameters
   In the first day of the 3rd and 6th week morning, selected 3 broilers from each replicate randomly, prepared heart blood, make serum samples, test the contect of Lysozyme and the activity of total protein (TP), total cholesterol (TC), glucose (Glu) and aspartate transaminase (GPT), glutamic pyruvic transaminase (GPT). Serum biochemical indicators were determined according to the kit instructions.
1.4 statistical method
  The test data were analyzed by SPSS10. 0.  Variance analysised and LSD multiple comparised by One-Way, and mark the result by Average±Standard error, and multiple comparison.
2 Results and Analysis
2.1 Effect of lysozyme, probiotics and Astragalus Polysaccharide on growth performance of Broilers
  Sheet 2 shows that: compared with the control group, supplement lysozyme, probiotics and astragalan into ration can improve broiler growth performance and feed conversion rate. Among them, compared to the control group, lysozyme increased 7.79%, the difference was significant, the Feed reward increase 3.93%, the difference was significant. Compared to astragalan, the weight of Lysozyme group increased 2.43%, the feed reward increased 2.86%. Compared with the control group, the weight of probiotic group increased of 6.55%, the difference was significant; Compared with the control group, the weight of astragalan increased of 5.25%, the difference was significant.
2.2 Effect of lysozyme, probiotics and Astragalus Polysaccharide on the immune function of Broilers
   Sheet 2 shows that: Supplement lysozyme into the early growth broiler's ration could improve the content of lysozyme in serum of broilers, compared to the control group, the content of lysozyme in serum increased 43. 16%, difference is very significant; Compared to the probiotic and and astragalan group,increased 34. 36% and 22. 84%, the difference was significant. Lysozyme could increase the total protein content in serum, compared with the control group, increased 41.58%, the difference was significant. Probiotics and astragalan can reduce the content of serum glucose and alanine aminotransferase effectively, compared with the control group, reduced 10. 93%,10. 98% and 38. 71%, 35. 75% respectively, the difference was significant.
Sheet 4 shows that: compared with the other groups, supplement lysozyme into the ration can improve the serum lysozyme content of broilers, increased by 33. 3% (P < 0. 05), 21. 5% (P < 0. 05) and 15. 8% (P > 0. 05); Reduce the content of cholesterol in serum, reduced by 11. 4% (P < 0. 05) and 8% (P < 0. 05) and 7. 9% (P< 0.05). Supplemente with lysozyme, probiotics and astragalus polysaccharide could increased the content of total protein, reduce the content of serum glucose and glutamic pyruvic transaminase in serum.
3 Conclusion
   Supplement  lysozyme and probioticsy etc into the ration of young broiler could  improve the growth performance, especially inprove the early growth performance; they also have different degrees impact on blood biochemical indexes. The weight gain and feed rward of lysozyme group were higher than in the other experimental groups, differences are significant; At the same time, lysozyme can  improve broiler Serum Lysozyme and total protein content significantly, improve the immune function.
